Changes induced by co-inoculation in nitrogen–carbon metabolism in cowpea under salinity stress BJM
Santos,Alexandra de Andrade; Silveira,Joaquim Albenísio Gomes da; Guilherme,Eliezer de Araujo; Bonifacio,Aurenivia; Rodrigues,Artenisa Cerqueira; Figueiredo,Márcia do Vale Barreto.
ABSTRACT To mitigate the deleterious effects of abiotic stress, the use of plant growth-promoting bacteria along with diazotrophic bacteria has been increasing. The objectives of this study were to investigate the key enzymes related to nitrogen and carbon metabolism in the biological nitrogen fixation process and to elucidate the activities of these enzymes by the synergistic interaction between Bradyrhizobium and plant growth-promoting bacteria in the absence and presence of salt stress. Cowpea plants were cultivated under axenic conditions, inoculated with Bradyrhizobium and co-inoculated with Bradyrhizobium sp. and Actinomadura sp., Bradyrhizobium sp. and Bacillus sp., Bradyrhizobium sp. and Paenibacillus graminis, and Bradyrhizobium sp. and...

Differential capacity of nitrogen assimilation between apical and basal leaf portions of a tank epiphytic bromeliad Braz. J. Plant Physiol.
Takahashi,Cassia A.; Ceccantini,Gregório C.T.; Mercier,Helenice.
The leaf is the main organ of the vegetative body of tank epiphytic bromeliads, which may be subdivided into apical and basal parts. Little is known about the existence of morphological, anatomical or physiological differences between these two leaf portions. The objective of this study was to verify the existence of a differential capacity to assimilate nitrogen from urea between basal and apical leaf portions of Vriesea gigantea, a tank epiphytic bromeliad. Plants were cultivated in vitro in the presence of 5 mM urea for 15 days. During this period, the activities of the enzymes glutamine synthetase (GS, EC 6.3.1.2) and NADH-dependent glutamate dehydrogenase (NADH-GDH, EC 1.4.1.2) were quantified, as well as the densities of trichomes and stomata found...

DROUGHT-INDUCED EFFECTS AND RECOVERY OF NITRATE ASSIMILATION AND NODULE ACTIVITY IN COWPEA PLANTS INOCULATED WITH BRADYRHIZOBIUM SPP. UNDER MODERATE NITRATE LEVEL BJM
Silveira,Joaquim Albenísio Gomes da; Costa,Roberto Cezar Lobo da; Oliveira,José Tadeu Abreu.
This study was carried out to establish comparative effects of drought and recovery on the nitrate assimilation and nodule activity related to N2 fixation in cowpea plants [Vigna unguiculata L. (Walp.)] previously inoculated with Bradyrhizobium spp. BR-3256 (CB-756) strain in the presence of 5 mol m-3 NO-3. Twenty-eight-day-old nodulated plants were submitted to water deprivation during 4 consecutive days and afterwards resupplied with nutrient solution during 2 days. The water deprivation caused a rapid increase in the nitrate content in root and a marked reduction in leaf nitrate reductase (NR) activity. In contrast nodule NR activity was slightly increased by water deprivation. Concomitantly, in nodules of water stressed plants, leghemoglobin and...

This work was aimed at locating Eucalyptus ESTs corresponding to the GS enzyme (Glutamine Synthetase, EC = 6.3.1.2) and to the D1 protein, which are directly related to resistance to herbicides that promote oxidative stress. Glutamine Synthetase corresponds to the site of action of the herbicide glufosinate. Herbicides that belong to groups such as ureas, uracils, triazines and triazinones act on the D1-Qb complex (receptor of electrons from the Photosystem II) by inactivating it. The clusters EGEQRT3302E01.g, EGEQRT3001F12.b; EGEZLV1203B04.g; EGBGFB1211H06.g and EGEZLV1205F09.g enclosed complete sequences (with 356 amino acids) of the Glutamine Synthetase enzyme. The cluster EGEQSL1054G06.g is a consensus of four reads and enclosed a complete sequence of...

Genetic polymorphism of glutamine synthetase and delta-9 desaturase in families of Pacific oyster Crassostrea gigas and susceptibility to summer mortality ArchiMer
David, Elise; Boudry, Pierre; Degremont, Lionel; Tanguy, Amelie; Quere, N; Samain, Jean-francois; Moraga, D.
Large-scale mortality events have been observed in Pacific oyster Crassostrea gigas on the west coast of France since the early 1980s, particularly during summer. In order to understand the causes of this mortality, two generations of oysters from single-pair matings were studied in three sites on the French Atlantic coast (Baie-des-Veys, Auray and Ronce-les-Bains). The present paper examines the role of two candidate genes in the susceptibility of oysters to summer mortality, and the selective pressure exerted by such mortality on their polymorphism. Glutamine synthetase (amino-acid metabolism) and delta-9 desaturase (lipid metabolism) genes were studied in the successive generations, using polymerase chain reaction single-strand conformation polymorphism...

Glial cell activity is maintained during prolonged inflammatory challenge in rats BJMBR
Borges,B.C.; Rorato,R.; Antunes-Rodrigues,J; Elias,L.L.K..
We evaluated the expression of glial fibrillary acidic protein (GFAP), glutamine synthetase (GS), ionized calcium binding adaptor protein-1 (Iba-1), and ferritin in rats after single or repeated lipopolysaccharide (LPS) treatment, which is known to induce endotoxin tolerance and glial activation. Male Wistar rats (200-250 g) received ip injections of LPS (100 µg/kg) or saline for 6 days: 6 saline (N = 5), 5 saline + 1 LPS (N = 6) and 6 LPS (N = 6). After the sixth injection, the rats were perfused and the brains were collected for immunohistochemistry. After a single LPS dose, the number of GFAP-positive cells increased in the hypothalamic arcuate nucleus (ARC; 1 LPS: 35.6 ± 1.4 vs control: 23.1 ± 2.5) and hippocampus (1 LPS: 165.0 ± 3.0 vs control: 137.5...

Physiological and N2-fixation-related traits for tolerance to drought in soybean progenies PAB
Cerezini,Paula; Kuwano,Biana Harumi; Neiverth,Walkyria; Grunvald,Anna Karolina; Pípolo,Antonio Eduardo; Hungria,Mariangela; Nogueira,Marco Antonio.
Abstract: The objective of this work was to evaluate six soybean (Glycine max) genotypes for physiological traits and biological nitrogen fixation in drought conditions, and their capacity for recovery after rewetting, based on yield components. The genotypes evaluated in a greenhouse were the following: 'BRS 317', susceptible to drought; R01-581F and R02-1325, which show biological nitrogen fixation tolerant to drought; and the BRB14-207525, BRB14-207526, BRB14-207527 breed lines resulting from crossings between commercial genotypes and PI 471938, which shows a slow-wilting phenotype under drought. Drought conditions were applied to the genotypes at the full-flowering stage (R2) for ten days, whereas control plants were kept well-watered. Photosynthetic...

SENSITIVITY OF MORNINGGLORY PLANTS AND THEIR PROGENIES TO GLUFOSINATE AMMONIUM Planta Daninha
BRITO,I.P.F.S.; MORAES,C.P.; MARCHESI,B.B.; CARBONARI,C.A.; VELINI,E.D..
ABSTRACT The sensitivity of weed species to herbicides is variable, and the behavior can be repeated or not in the next generation. The objective of this study was to evaluate the response of morningglory plants to different doses of glufosinate ammonium and the sensitivity of plant populations and their progenies to the herbicide. Three studies were conducted, all in a greenhouse and repeated in two periods. In the first study, two experiments were conducted to examine the dose-response curve, and the treatments were seven different doses of the herbicide glufosinate ammonium (0, 50, 100, 200, 400, 800, and 1,600 g a.i. ha-1), with four replications each. In the second study, which examined the range in sensitivity of morningglory to glufosinate ammonium,...
